Generation and Screening of Ditopic Dynamic Combinatorial Libraries

چکیده
Dynamic Combinatorial Chemistry (DCC) is a new supramolecular concept that extends beyond static combinatorial chemistry towards adaptive chemical systems. This concept relies on reversible interchange between sets of basic components to generate continually interconnecting adducts, giving access to dynamic combinatorial libraries (DCLs) comprising all possible combinations of the components available. Such libraries allow for the target-driven generation of the active constituent(s) of the libraries, thus performing a selfscreening process by which the active species are preferentially expressed and retrieved from the DCL. In this study, the implementation of such libraries on biological interactions was examined and dynamic libraries, generated from small arrays of initial precursors, were probed against binding to biological receptors.

Assessment of "drug-likeness" of a small library of natural products using chemoinformatics
Even though natural products has an excellent record as a source for new drugs, the advent of ultrahigh-throughput screening and large-scale combinatorial synthetic methods, has caused a decline in the use of natural products research in the pharmaceutical industry. This is due to the efficiency in generating and screening a high number of synthetic combinatorial compounds; whereas traditional ...

متن کاملReversible Sulfur Reactions in Pre-Equilibrated and Catalytic Self-Screening Dynamic Combinatorial Chemistry Protocols
Dynamic Combinatorial Chemistry (DCC) is a recently introduced supramolecular approach to generate dynamically interchanging libraries of compounds. These libraries are made of different building blocks that reversibly interact with one another and spontaneously assemble to encompass all possible combinations. If a target molecule, for instance a receptor is added to the system and one or more ...
متن کاملMixture-based combinatorial libraries from small individual peptide libraries: a case study on α1-antitrypsin deficiency.
The design, synthesis and screening of diversity-oriented peptide libraries using a "libraries from libraries" strategy for the development of inhibitors of α1-antitrypsin deficiency are described. The major buttress of the biochemical approach presented here is the use of well-established solid-phase split-and-mix method for the generation of mixture-based libraries. The combinatorial techniqu...
متن کاملTwo-vial, LC-MS identification of ephedrine receptors from a solution-phase dynamic combinatorial library of over 9000 components.
Reports on dynamic combinatorial chemistry have almost exclusively involved small libraries of 10-100 compounds. We now show how more than 9000 compounds can be screened in a single LC-MS analysis to reveal a series of new receptors that bind ephedrine in water. These results demonstrate the feasibility of screening DCLs that are substantially larger than the solution-phase libraries reported t...
